目的

分离、鉴定铁皮石斛叶中的黄酮碳苷类成分,并评价其体外抑制
α
-葡萄糖苷酶的活性。
方法

采用大孔吸附树脂和制备型高效液相色谱法对铁皮石斛叶中的黄酮碳苷类成分进行分离和纯化,通过紫外光谱、核磁共振、高分辨电喷雾离子化质谱等波谱方法对化合物进行推导和鉴定。考察黄酮碳苷类成分与阳性对照(阿卡波糖)的
α
-葡萄糖苷酶体外抑制活性。
结果

从铁皮石斛叶中分离得到5个芹菜素-6,8-
C
-二糖苷,鉴定为芹菜素-6
-C-α
-L-鼠李糖-8
-C-β
-D-异鼠李糖(
1
)、夏佛塔苷(
2
)、异夏佛塔苷(
3
)、异佛莱心苷(
4
)和佛莱心苷(
5
)。化合物
1
~
5
和阿卡波糖抑制
α
-葡萄糖苷酶的半数抑制浓度分别为(1.79±1.27)、(2.05±0.72)、(1.93±0.67)、(1.09±0.46)、(1.36±0.58)、(18.69±1.24)μmol/L。
结论

从铁皮石斛叶中分离得到5个具有
α
-葡萄糖苷酶抑制活性的芹菜素-6,8-
C
-二糖苷类成分,其中化合物

为新化合物,化合物

首次从该植物中分离得到。
OBJECTIVE
2
To separate and identify the flavone
C
-glycosides from the leaves of
Dendrobium officinale
, and to evaluate their
in vitro
inhibitory activities to α-glucosidase.
METHODS
2
The flavone
C
-glycosides from the leaves of
D. officinale
were separated and purified by macroporous adsorption resin and preparative high-performance liquid chromatography. The structure of obtained compound was elucidated and identified by spectroscopic methods, such as ultraviolet spectrum, nuclear magnetic resonance, high-resolution electrospray ionization mass spectrometry, etc. The
in vitro
inhibitory activities of flavone
C
-glycosides and positive control (acarbose) to
α
-glucosidase were investigated.
RESULTS
2
Five apigenin-6,8-di-
C
-glycosides were isolated and purified from the leaves of
D. officinale
, and identified as apigenin-6-
C
-
α
-L-rhamnosyl-8-
C
-
β
-D-quinovoside(
1
), schaftoside (
2
),isoschaftoside(
3
),isoviolanthin(
4
) and violanthin(
5
). Half inhibitory concentration of compound
1
-
5
and acarbose inhibiting
α
-glucosidase were (1.79±1.27),(2.05±0.72),(1.93±0.67),(1.09±0.46),(1.36±0.58),(18.69±1.24) μmol/L, respectively.
CONCLUSIONS

Five apigenin-6,8-di-
C
-glycosides with
α
-glucosidase inhibitory activity are isolated from the leaves of
D. officinale
, of which compound

is a new compound and compound
2
is isolated from this plant for the first time.
