核因子E2相关因子2(Nrf2)/谷胱甘肽过氧化酶4(GPX4)信号通路在肿瘤的发生发展中扮演着关键角色,其参与了肿瘤细胞的增殖、凋亡、铁死亡、侵袭、迁移以及耐药等过程。本文综述了中药通过调控Nrf2/GPX4信号通路抗肿瘤的研究,结果发现,黄酮类(银杏素、木犀草素等)、萜类(苍术素、葫芦素B等)、皂苷类(重楼皂苷Ⅰ、重楼皂苷Ⅶ)、酯类(鸦胆子苦醇)等中药有效成分、中药提取物(消瘤藤总香豆素、湖北枫杨总黄酮)以及中药复方(附芍地芩方、消癌解毒方等)能够通过抑制Nrf2/GPX4信号通路及其上下游因子蛋白的表达,提高Fe
2+
、脂质过氧化等来促进肿瘤细胞铁死亡,进而发挥抗肿瘤作用。
Nuclear factor-erythroid 2-related factor 2 (Nrf2)/glutathione peroxidase 4 (GPX4) signaling pathway plays a key role in the occurrence and development of tumors, and is involved in tumor cell proliferation, apoptosis, ferroptosis, invasion, migration, and drug resistance. Based on the Nrf2/GPX4 signaling pathway, this paper summarizes the research progress of the anti-tumor effects of traditional Chinese medicine. It is found that flavonoids (ginkgetin, luteolin, etc.), terpenoids (atractylenol
ide, cucurbitacin B, etc.), saponins (polyphyllin Ⅰ, polyphyllin Ⅶ), ester (brusatol) and other effective components, and traditional Chinese medicine extracts (total coumarins in
Pileostegia tomentella
and total flavonoids of
Pterocarya hupehensis
Skan), traditional Chinese medicine compounds (Fushao diqin fang, Xiaoai jiedu fang, etc.) can promote ferroptosis in tumor cells by inhibiting Nrf2/GPX4 signaling pathway and the expressions of its upstream and downstream factor proteins, as well as by increasing Fe
2+
levels and lipid peroxidation, thereby exerting an antitumor effect.
