OBJECTIVE: To preliminarily investigate the constituents absorbed into the blood of ethanol extract from Mongolia medicine Scabiosa comosa in rats, and to provide reference for clarifying the effective substance basis of the medicine. METHODS: Ten rats were randomly divided into administration group (0.3 g/100 g by crude drug) and blank group (0.5% CMC-Na solution), with 5 rats in each group. The were given relevant medicine intragastrically 2 mL/100 g, at 8:00 morning and evening, for consecutive 3 d. 30 min after last administration, blood samples were collected from hepatic portal vein. The containing serum samples and blank serum samples were prepared by organic solvent precipitation. UPLC-MS/MS method was adopted by using S. comosa as control. Through comparing chemical components of 3 kinds of samples, constituents of ethanol extract from S. comosa in rats after intragastric administration were identified on the basis of compound control and literature data. RESULTS: Totally 39 constituents absorbed into blood were found, of which 17 were prototype components and 22 related metabolites. The prototype components were mainly flavonoids and phenylpropionic acid. Methylation, sulfation and glucose aldehyde reactions were observed in rats. CONCLUSIONS: The constituents into the blood of ethanol extract from S. comosa in rats are confirmed preliminarily after intragastric administration, which lay a foundation for further metabolism study in vivo.
