OBJECTIVE: To optimize the extraction technology of Wikstroemia indica. METHODS: Using the active ingredient contents of umbelliferone and daphnoretin, and extraction yield as comprehensive evaluation indexes, solvent dosage, ethanol volume fraction, percolation rate as investigation factors, single factor test was conducted to compare the 3 extraction methods (percolation method, boiling method, reflux extraction method), and screen the optimized one; and the technology conditions of optimized method were optimized and verified. RESULTS: The comprehensive score ordered as percolation method, reflux extraction method, boiling method, which was better by percolation method. The optimal technology conditions was as follow as percolation solvent of 70% ethanol, 14-fold herbs at percolation rate of 5 mL/(min·kg); the comprehensive scores in 3 tests were 98.3, 99.3, 98.8 (RSD=0.51%, n=3). CONCLUSIONS: Percolation method is optimized for extracting active ingredient of W. indica, which is stable and reproducible.
