目的:比较巴戟天及其不同炮制品对脾肾阳虚模型大鼠的改善作用。方法:将80只大鼠随机分为正常组(生理盐水)、模型组(生理盐水)、辅料组(甘草汁,0.063 g/kg)、全鹿丸组(阳性药物,1.89 g/kg)和巴戟天、巴戟肉、盐巴戟、制巴戟组[给药剂量均为1.05 g(生药)/kg]。除正常组外,其余各组大鼠均采用ig大黄水煎液+im氢化可的松复制脾肾阳虚模型,并同时ig相应药物,每天1次,连续15 d。测定血清中睾酮(T)、雌二醇(E2)、环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、三碘甲状腺原氨酸(T3)、甲状腺素(T4)、促肾上腺素皮质激素(ACTH)水平,并计算E2/T、cAMP/cGMP比值。结果:与正常组比较,模型组大鼠血清中T 、T3、T4、cGMP水平降低,E2、cAMP、ACTH水平及E2/T、cAMP/cGMP比值升高(P<0.05或P<0.01)。与模型组比较,各给药组大鼠血清中T水平升高,E2、cAMP、ACTH水平及cAMP/cGMP比值降低(P<0.05或P<0.01),且以盐巴戟组大鼠血清中T水平最高、E2/T比值最低,制巴戟组大鼠血清中E2、cAMP、ACTH水平及cAMP/cGMP比值最低;巴戟天组和制巴戟组大鼠血清中cGMP水平升高(P<0.01);巴戟天组、盐巴戟组和制巴戟组大鼠血清中T3水平及各给药组大鼠血清中T4水平升高(P<0.05或P<0.01),且以制巴戟组升高最为显著。结论:在巴戟天及其不同炮制品中,制巴戟对脾肾阳虚大鼠的改善作用更明显。
OBJECTIVE: To compare the improvement effects of Morinda officinalis and its processed products on rats with spleen-kidney Yang deficiency. METHODS: 80 rats were randomly divided into normal group (normal saline), model group (normal saline), excipients group (succus liquiritiae, 0.063 g/kg), Quanlu pill group (positive drug, 1.89 g/kg), M. officinalis group, M. officinalis without wood, M. officinalis processed by salt and M. officinalis processed by licorice-juice groups [1.05 g (crude drug)/kg]. Those groups were given Rheum palmatum water solution intragastrically and hydrocortisone intramuscularly to induce spleen-kidney Yang deficiency model, and given relevant medicine intragastrically once a day for consecutive 15 d. The levels of testosterone(T), estradiol(E2), cAMP, cGMP, triiodothyronine (T3), thyroxine (T4), adrenal cortex hormone(ACTH) in the serum were measured, and E2/T and cAMP/cGMP were calculated. RESULTS: Compared with normal group, the serum level of T, T3, T4 and cGMP decreased in model group, while the levels of E2, cAMP and ACTH, E2/T and cAMP/cGMP increased (P<0.05 or P<0.01). Compared with model group, serum level of T increased in treatment groups, while serum levels of E2, cAMP, ACTH and cAMP/cGMP decreased (P<0.05 or P<0.01); serum level of T was highest and E2/T was the lowest in M. officinalis processed by salt group; serum level of E2, cAMP, ACTH and cAMP/cGMP were the lowest in M. officinalis processed by licorice-juice group; serum level of cGMP increased in M. officinalis group and M. officinalis processed by licorice-juice group (P<0.01); serum levels of T3 in M. officinalis, M. officinalis processed by salt and M. officinalis processed by licorice-juice groups, and T4 increased in all treatment groups (P<0.05 or P<0.01), especially in M. officinalis processed by licorice-juice group. CONCLUSIONS: M. officinalis processed by licorice-juice is better in improving spleen-kidney Yang deficiency, compared to M. officinalis and its other processed products.
